In Vitro Culture
Short-term explant cultures derived from flank tumor xenograft tissue can be used for in vitro cell culture-based assays. The Mayo Clinic Brain Tumor Patient-Derived Xenograft National Resource typically doesn't keep cells in culture for more than three to four weeks.
Growth characteristics of each patient-derived xenograft as an explant culture can vary in fetal bovine serum (FBS)-containing media or neural basal stem cell media.
Most lines are initially plated on tissue culture plates coated with Matrigel (FBS cultures) or laminin (neural stem cell culture) and subsequently passaged into plates without coatings.
