About this study

The purpose of this study is to evaluate the safety and efficacy of a single dose of ex vivo base-edited, autologous CD34+ human hematopoietic stem and progenitor cells (HSPCs) in patients with SCD and severe VOCs. Like allogenic HSCT, these therapies are designed to replace the patient’s bone marrow (BM) with hematopoietic stem cells (HSCs) that encode nonsickling globins. However, as an autologous therapy, BEAM-101 (the IMP in this study) has the potential advantages of eliminating the morbidity and mortality of graft-versus-host disease (GVHD) and greatly broadening access to the majority of patients who lack a matched sibling donor.

BEAM-101 utilizes base editing to introduce specific point mutations that are designed to induce the expression of therapeutic, nonsickling hemoglobin (Hb). BEAM-101 introduces point mutations in the HBG1 and HBG2 promoters, which mimic naturally occurring mutations that disrupt binding of B-cell lymphoma/leukemia 11A gene (BCL11A), a transcriptional repressor of fetal hemoglobin (HbF). Individuals with these mutations have the condition known as hereditary persistence of fetal hemoglobin (HPFH) and continue to have nonsickling HbF expression throughout their life.

About this study

The purpose of this study is to obtain safety, efficacy, and pharmacodynamic data on the use of vutrisiran in patients with ATTR amyloidosis with cardiomyopathy who continued on extended use of vutrisiran, or switched from patisiran.

About this study

This study will test a study drug called cemiplimab to see if it can help treat early-stage cutaneous squamous cell carcinoma (CSCC), a type of skin cancer. Cemiplimab works by helping the immune system to kill cancer cells. It binds to a protein called programmed cell death-1 (PD-1) on the surface of certain immune cells. The main purpose of this study is to compare how well cemiplimab works compared to surgery, when injected into the lesion.

About this study

This pilot, two-arm, randomized study will assess the feasibility of measuring the efficacy of aspirin, compared with observation, for reducing PGE2 and a subset of factors in paired biopsies of endometrium tissue from post-menopausal women. Eligible participants will undergo two endometrial biopsies (i.e. baseline, week-6, or sooner if chosen through shared decision making) separated by 6 weeks (+/- 7 days).

At both timepoints, blood and urine will also be collected to investigate mechanisms of tissue changes. As secondary aims, additional biomarker testing of paired biospecimens (urine, blood, and tissue) will be conducted to assess the correlation of results with treatment group, and clinicopathologic status of the endometrium (i.e. PMB and endometrial thickening). Also, correlations between collected urine, blood, and tissue measures will be explored.

Based on prior knowledge that age and body composition play a role in mechanisms related to endometrial status, sub-group analyses of the primary and secondary endpoints will be conducted. Additional sub-groups in race and ethnicity will be explored as the data allow, due to known differences in endometrial cancer mortality.

Finally, qualitative interviews will be utilized to investigate feasibility of future clinical trials in the community.
