Equipment and Facilities
The Metabolomics Nuclear Magnetic Resonance Spectroscopy (NMRS) will be integrated with the NMR Facility and use facility equipment. The NMR Facility offers five high-field NMR spectrometers for data acquisition, computers for NMR data analysis and a fully equipped wet laboratory for NMR sample preparation:
- Bruker Avance III 600 and 700 MHz spectrometers are used for obtaining high-resolution 1H NMR spectra from tissue extracts and body fluids to quantitate metabolite levels.
- A Bruker 500 MHz and wide bore 700 MHz NMR spectrometer (Avance) is used for 13C and 18O-assisted 31P NMR studies to determine the levels and dynamics of cellular carbon- and phosphoryl-containing metabolites.
- A high-sensitivity 600 MHz BB cryoprobe for 31P and 13C NMR, a high-throughput sample handler for 1H NMR metabolomics, and 1.7 mm triple resonance NMR MicroCryoProbe for metabolite identification will be acquired to expand core analytical capacity.
Two NMR spectrometers (the 300 MHz and 700 MHz wide bore) are equipped with mini- and micro-imaging accessories that enable the recording of high-resolution NMR spectra of metabolites in small animals, perfused organs, tissues and cell cultures. The 300 MHz spectrometer has a magic-angle spinning probe for high-resolution multinuclear spectroscopy of metabolites present in soft solids such as tissue and cell pellets.
The Metabolomics NMRS Core is also equipped for stable isotope labeling, organ perfusion systems, cell culture systems and whole blood metabolic labeling tools for teaching purposes.
The Metabolomics NMRS Core laboratory will utilize a Columbus Instruments' mouse Metabolic Modular Treadmill System for stress testing of transgenic animals.
