Centrosome Abnormalities in Plasma Cell Neoplasm

As myeloma is characterized by widespread aneuploidy, we are interested in studying possible mediating mechanisms. One possible mechanism is centrosome amplification that has been described in other malignancies and shown to be associated with aneuploidy. We showed that centrosome amplification (E and F) is already present at the pre-malignant MGUS stage and increases with progression to MM. In addition, we derived a gene-expression based 3-gene index (centrosome index) that correlated strongly with centrosome amplification and identify a group of patients with very poor prognosis regardless of treatment (Chng et al. Blood 2006; 107: 3669-3675).

We are now defining the unique molecular profile of patients with high centrosome index in order to identify novel ways of treating these very poor prognosis patients.
